A rabbit polyclonal antibody from Antibodies.com that simultaneously recognises the activating phosphorylation sites of three p21-activated kinase family members — PAK1 at threonine 423, PAK2 at threonine 402, and PAK3 at threonine 421. These conserved residues sit within the kinase activation loops, and their phosphorylation reflects the catalytically engaged state of each enzyme, so a single reagent can be used to monitor pathway activation across the trio without the need for three separate primaries. The antibody is raised in rabbit, purified as a polyclonal IgG preparation, and supplied unconjugated at 200 µl per vial, leaving the conjugate choice open for downstream detection chemistry. Validated applications are immunohistochemistry (IHC) and immunofluorescence (IF), and the reagent has been tested for reactivity against human, mouse, and rat samples, making it useful for cross-species studies in fixed tissue and cultured cells. Typical uses include profiling PAK signalling in cancer invasion and migration models, neuronal development and cytoskeletal-remodelling research, and investigations of Rho-family GTPase effector pathways where coordinated PAK1/2/3 activation is of interest. Because it is a polyclonal, the reagent tolerates minor sequence variation around the conserved phospho-threonine motif and provides redundancy against the modest species differences between the three PAK paralogues, while the unconjugated format allows pairing with HRP-, biotin-, or fluorophore-labelled secondaries depending on the assay design.
