A rabbit polyclonal IgG antibody raised against the phosphorylated activation-loop threonines of the PAK family of serine/threonine kinases. The Antibodies.com reagent detects phosphorylated PAK1 (Thr423), phosphorylated PAK2 (Thr402), and phosphorylated PAK3 (Thr421), the conserved residues whose autophosphorylation marks the fully activated state of each kinase isoform. Supplied as an unconjugated preparation in a 100 µl volume, it is intended for use in immunohistochemistry (IHC) and immunofluorescence (IF) workflows on human, mouse, and rat samples. This combined specificity makes the antibody especially useful for interrogating PAK pathway activity in tissue sections and fixed-cell preparations, where parallel readouts of PAK1, PAK2, and PAK3 activation allow direct comparison of isoform engagement in a single staining experiment.

PAK1, PAK2, and PAK3 are downstream effectors of Rho-family GTPases such as Cdc42 and Rac, and phosphorylation of the threonine in the activation loop is widely used as a read-out of receptor-driven and cytoskeletal signaling. Because the antibody recognizes all three phosphorylation sites on the corresponding residue, it is well suited to studies of tumor sections, neuronal tissue, and developmental models where simultaneous assessment of PAK isoform activation informs questions about cell migration, proliferation, and cytoskeletal remodeling. It is provided as a research-use-only polyclonal reagent that performs on standard formaldehyde-fixed, paraffin-embedded material as well as on immunofluorescence preparations of cultured cells, giving laboratories flexibility to address PAK pathway biology across both archival and freshly fixed specimens.
