Raised against peptidyl arginine deiminase type 4 (PADI4), this rabbit polyclonal IgG reagent from Antibodies.com supports research into calcium-driven citrullination, a post-translational modification in which arginine residues on histones H2A, H3, and H4 are converted to citrulline. PAD4 activity weakens electrostatic contacts between histones and DNA, driving chromatin decondensation and the extrusion of neutrophil extracellular traps (NETs) during innate immune responses. Aberrant citrullination has been linked to rheumatoid arthritis, multiple sclerosis, and tumor microenvironments, and neutrophils lacking PAD4 fail to citrullinate histones or release NETs in response to stimuli such as ionomycin, LPS, and PMA.

Validated for western blot, immunohistochemistry, and immunofluorescence workflows, the antibody cross-reacts with human, mouse, and rat samples, enabling comparative studies across model organisms. It is supplied as 200 µl of unconjugated liquid reagent in a vial format, intended for research use only. Typical applications include tracking PADI4 expression in neutrophils, profiling histone citrullination in synovial tissue, and characterizing NET-associated antigens targeted by anti-citrullinated protein antibodies in autoimmune disease models.
