Raised in rabbit as a polyclonal IgG, this unconjugated primary reagent from Antibodies.com targets PADI4, a calcium-dependent enzyme that converts arginine residues to citrulline on histones H3 (notably at Arg-8 and Arg-17) and histone H4 (at Arg-3), as well as on non-histone substrates such as EP300/P300. The antibody detects the approximately 73.9 kDa protein in cell and tissue samples and is supplied at a 100 µL volume, validated for Western blot, immunohistochemistry, and immunofluorescence workflows across human, mouse, and rat samples. Researchers apply it to compare enzyme abundance between disease and normal specimens in studies of citrullination-driven chromatin remodelling, neutrophil extracellular trap (NET) formation, rheumatoid arthritis synovial tissue, multiple sclerosis lesions, and solid tumour biology. Because PADI4 activity is commonly inferred from citrullinated histone H3 staining on matched sections, results obtained with this reagent are typically correlated with citrulline-specific readouts in the same workflow. The unconjugated IgG format allows flexible pairing with user-selected secondary antibodies, fluorophores, or HRP conjugates, supporting both chromogenic immunohistochemistry on paraffin-embedded tissue and immunofluorescent labelling of cultured cells or frozen sections. To preserve activity across multiple freeze-thaw cycles, the preparation should be stored as aliquots at low temperature and protected from light. The reagent is intended for research use only.
