The Antibodies.com polyclonal antibody selectively recognizes the phosphorylated Ser352 residue within the activation loop of the 90 kDa ribosomal S6 kinase (P90RSK, also referred to as RSK1 or RPS6KA1), a serine/threonine kinase that operates downstream of the Ras/Raf/MEK/ERK signaling cascade. Phosphorylation at Ser352 is one of the activating modifications catalysed by ERK1/2 and contributes to full kinase activity, making this reagent a useful marker for studies of growth factor, mitogen, or oncogenic pathway activation in human cells and tissues.

Provided as 50 µl of unconjugated rabbit IgG, the reagent is suited to Western blot detection of phosphorylated P90RSK in human samples. Researchers commonly use such phospho-specific antibodies to monitor pathway engagement after stimulation with EGF, FGF, PDGF, or serum, and to compare activation states between treated and untreated controls, including work with MEK or ERK pathway inhibitors. The antibody is raised in rabbit, purified as a polyclonal IgG, and is intended for research use only in vitro.

For reliable performance, aliquot upon arrival, store at -20 °C, and limit repeated freeze-thaw cycles to preserve phospho-specific reactivity. Lysis buffers should be supplemented with phosphatase inhibitors so the Ser352 phosphorylation mark is retained in the prepared lysate prior to SDS-PAGE and immunoblotting.
