Phosphorylation at Ser20 disrupts p53 binding to its negative regulator MDM2, leading to rapid stabilization, nuclear accumulation, and transcriptional activation of downstream targets such as p21, Bax, and PUMA, which govern cell cycle arrest and apoptosis.
