p14 ARF is the alternative reading frame product of the INK4a/ARF locus and acts through the p53 stress pathway by binding MDM2, blocking MDM2-mediated ubiquitination of p53, and stabilizing p53 to drive cell cycle arrest, senescence, or apoptosis. Loss of p14 ARF expression, frequently through CDKN2A homozygous deletion or promoter methylation, is reported across melanoma, glioblastoma, non-small cell lung carcinoma, pancreatic adenocarcinoma, and mesothelioma, making it a widely used reference marker in tumor profiling studies.

Researchers commonly apply this reagent to confirm ARF status in cell line panels including ARF-deficient and reconstituted controls, to assess nucleolar localization by immunofluorescence, and to detect the ~14 kDa band in western blot lysates.
