Affinity-purified from rabbit serum and supplied as 50 µl of liquid polyclonal IgG, this unconjugated Antibodies.com reagent specifically recognizes the N-terminal region of OCRL inositol polyphosphate-5-phosphatase and is validated for Western blot detection across human, mouse, and rat samples. Because the antibody is delivered without a fluorophore or enzyme tag, it pairs cleanly with HRP-conjugated secondary reagents for chemiluminescent blots or with fluorescent secondaries for multiplex imaging, and it is formulated without azide so it does not interfere with downstream coupling chemistries.

OCRL is a multi-domain X-linked lipid phosphatase built around a central inositol 5-phosphatase catalytic module flanked by an ASH domain and a RhoGAP-like domain, with an N-terminal PH domain that connects through a flexible linker carrying an AP-2-binding FxDxF motif. The enzyme hydrolyzes phosphatidylinositol 4,5-bisphosphate and PI(3,4,5)P3 at the trans-Golgi network, plasma membrane, and clathrin-coated endocytic pits, recruiting to those sites through two clathrin-box motifs and through interactions with the small GTPases Rac, Cdc42, and Rab5 plus the endocytic adaptor APPL1. Loss-of-function variants cause Lowe syndrome (oculocerebrorenal syndrome) and Dent disease type 2, conditions marked by congenital cataracts, intellectual disability, proximal tubule Fanconi-type reabsorption defects, neonatal hypotonia, defective megalin recycling, accumulation of PtdIns(4,5)P2 on early endosomes, and failure of primary cilium formation in patient fibroblasts.

Researchers use this primary antibody to profile OCRL expression, follow its subcellular redistribution, and probe phosphoinositide signaling in patient-derived fibroblasts, OCRL-knockout cell lines, renal proximal tubule epithelial models, and neuronal cultures. Recommended Western blot dilution is 1:500 to 1:2,000, where the antibody labels a band near the 104 kDa expected molecular weight of full-length OCRL. For storage, keep the vial frozen at -20°C, divide into smaller working aliquots to limit freeze-thaw cycles, and briefly centrifuge to clear any minor precipitate that may form during refrigeration; the reagent is intended for research use only.
