The NFκB-p65 subunit (RelA) is a central transcription factor in the canonical NF-κB signaling pathway, driving expression of genes involved in inflammation, immunity, and cell survival. Phosphorylation at threonine 429 (T429) is an activating post-translational modification that influences transcriptional activity and nuclear translocation. This polyclonal reagent from Antibodies.com is raised in rabbit and selectively detects the p65 subunit when phosphorylated at T429, enabling researchers to monitor pathway activation across stimulated and unstimulated samples.

Validated for Western blot and immunohistochemistry, the antibody reacts with human, mouse, and rat samples, making it broadly applicable to translational studies in cell lines, primary tissues, and animal disease models. IHC use allows visualization of phospho-p65 localization in fixed tissue sections, including tumor biopsies and inflammatory lesions, while Western blot detection provides quantitative readouts of pathway activation following cytokine, LPS, or stress stimulation.

The product is supplied as 50 µl of a 1 mg/ml stock in phosphate buffered saline (pH 7.2) containing 0.05% sodium azide as a preservative. Sodium azide is incompatible with some HRP-based detection chemistries, so an azide-free working buffer is recommended when developing with peroxidase substrates. The vial should be refrigerated and aliquoted upon receipt to limit freeze-thaw cycles if extended storage is needed.

This unconjugated polyclonal antibody integrates with both fluorescence and chromogenic detection schemes, allowing flexibility in multiplex staining panels or co-immunoprecipitation workflows.
