Nestin (NES) is a class VI intermediate filament protein widely used as a marker of neural stem and progenitor cells in the developing central nervous system, and as a marker of tumor-initiating populations in gliomas and other malignancies. This unconjugated rabbit polyclonal reagent from Antibodies.com targets the human NES protein and is validated for Western blot, immunohistochemistry, immunofluorescence, and flow cytometry workflows.

In Western blot, the antibody recognizes Nestin in lysates from neural progenitor cultures, stem-cell-derived rosettes, or glioma cell lines. Immunohistochemistry on fixed tissue sections reveals the characteristic filamentous cytoplasmic staining pattern in developing brain, subventricular zone, and tumor specimens where Nestin-positive cells reside. Immunofluorescence on fixed cultured cells highlights the cytoskeletal network, while flow cytometry supports quantification of Nestin-expressing populations among dissociated neural or tumor-derived spheres.

The reagent is supplied as 200 µl of polyclonal IgG in a vial, allowing working aliquots to be prepared to limit freeze-thaw exposure. Store refrigerated, protect from light, and handle under typical conditions for research use only. The polyclonal format recognizes several epitopes on the NES target, which can be useful when studying splice variants or post-translationally modified forms reported in neural stem cell and tumor biology.
