Anti-NCSTN Antibody

Nicastrin (NCSTN) is the single-pass type I transmembrane glycoprotein that acts as the scaffold subunit of the gamma-secretase complex, an intramembrane aspartyl protease composed together with presenilin (PSEN1/PSEN2), APH-1, and PEN-2. This rabbit polyclonal IgG primary antibody, supplied by Antibodies.com at 200 µL per vial in an unconjugated format, has been validated for Western Blot detection of endogenous Nicastrin in human and rat samples.

The mature Nicastrin protein carries extensive N-linked glycosylation, so the immature form migrates at approximately 110 kDa while the fully processed glycosylated form incorporated into active gamma-secretase runs at approximately 130 kDa. The gamma-secretase complex cleaves more than 140 type I transmembrane substrates, with the amyloid precursor protein (APP) and Notch receptors being the most studied: regulated intramembrane proteolysis of APP liberates amyloid-beta peptides that accumulate in senile plaques, and Notch cleavage releases the Notch intracellular domain for nuclear signaling.

NCSTN is widely interrogated in Alzheimer disease models that track amyloidogenic processing, in Notch-pathway studies of hematopoietic lineage decisions and developmental signaling, and in skin-biology work tied to familial acne inversa, in which loss-of-function mutations in NCSTN have been mapped. The unconjugated IgG format lets researchers pair the reagent with HRP-, fluorophore-, or biotin-tagged secondary antibodies, supporting chemiluminescent or fluorescent Western Blot workflows on PVDF or nitrocellulose membranes using standard lysate preparation.
