Affinity-purified from rabbit serum and raised against the human MutS homolog 2 (MSH2) mismatch repair protein, this polyclonal IgG reagent from Antibodies.com is supplied at 50 µL in an unconjugated format validated for western blot (WB) and immunoprecipitation (IP) workflows.
MSH2 is the keystone subunit of the post-replicative DNA mismatch repair (MMR) pathway. It pairs with MSH6 to form the MutSα heterodimer, which scans newly replicated DNA for single-base mismatches and small insertion-deletion loops, and pairs with MSH3 to form MutSβ, which recognizes larger loops up to about 13 nucleotides. In both complexes MSH2 contributes the ATPase activity that drives mismatch recognition and converts the heterodimer into a sliding clamp that diffuses along the DNA backbone. Germline loss-of-function mutations in MSH2 cause Lynch syndrome (HNPCC), and somatic inactivation drives the microsatellite instability seen in a substantial fraction of sporadic colorectal, endometrial, ovarian, and gastric tumors, making MSH2 a routine marker in tumor-typing immunohistochemistry and mismatch-repair-deficiency screening.
The reagent is reactive with Human, Mouse, and Rat samples. On western blot of whole-cell lysates it yields a band at the predicted molecular weight of 105 kDa, while the unconjugated IgG format leaves the end user free to pair it with any compatible secondary antibody or detection chemistry. For immunoprecipitation, the antibody pulls down endogenous MSH2 from whole-cell extracts, supporting downstream analysis of MutSα and MutSβ complex assembly by western blot or mass spectrometry.
For routine handling, aliquot the vial upon arrival and store at -20 °C to limit freeze-thaw cycles; short-term working stocks may be kept at 4 °C. This reagent is intended for research use only.
