Raised in rabbit as an unconjugated polyclonal IgG and supplied as a 50 µl liquid vial, this Antibodies.com reagent detects MRPS16, a 137-amino-acid component of the small 28S subunit of the human mitochondrial ribosome that is encoded at chromosomal locus 10q22.2. The precursor carries an N-terminal mitochondrial targeting peptide that is cleaved during mitochondrial import, leaving a mature peptide of about 11.5 kDa, while SDS-PAGE typically resolves the band near 15 kDa. The target sequence is more than 91% identical between mouse and human and is among the most conserved proteins of the mammalian mitoribosome. The reagent cross-reacts with the human, mouse, and rat antigen and is validated by the supplier for Western blot only. Researchers using this reagent to study mitochondrial ribosome assembly, intra-mitochondrial translation, or Combined Oxidative Phosphorylation Deficiency type 2 (COXPD2; OMIM 610498), an autosomal recessive disorder caused by nonsense variants such as R111X (rs104894168) in MRPS16, commonly pair it with mitochondrial fractionation, sucrose-gradient sedimentation of ribosomes, or co-immunoprecipitation from mitochondrial-enriched lysates. For storage, aliquot on receipt and store at -20 °C, protected from light; avoid repeated freeze-thaw cycles to preserve the affinity-purified IgG. For research use only.
