This rabbit polyclonal reagent from Antibodies.com targets matrix metalloproteinase 12 (MMP-12), also known as macrophage elastase or macrophage metalloelastase, a zinc-dependent endopeptidase that degrades elastin and other components of the extracellular matrix. The antibody was raised against a synthetic peptide corresponding to a region surrounding residue D411 of the human MMP-12 sequence and is supplied as an affinity-purified polyclonal reagent.

The product is provided as 50 µl of unconjugated antibody at approximately 1 mg/ml in phosphate buffered saline (PBS) containing 0.05% sodium azide at pH 7.2. It is validated for Western blot (WB) and immunohistochemistry (IHC) on human samples. As an unconjugated primary antibody, it can be paired with a user-selected secondary reagent such as HRP-conjugated anti-rabbit IgG for chemiluminescent immunoblotting on nitrocellulose or PVDF membranes, or with an enzyme- or fluorophore-labeled secondary for chromogenic detection on formalin-fixed paraffin-embedded tissue sections.

MMP-12 is widely investigated in the context of chronic obstructive pulmonary disease (COPD), emphysema, smoking-related lung injury, atherosclerosis, abdominal aortic aneurysm, wound healing, and inflammatory tissue remodeling, where its elastolytic activity contributes to matrix breakdown in affected tissues. The antibody recognizes the human MMP-12 protein and is used to detect endogenous expression in cell or tissue lysates by Western blot and to localize MMP-12 within tissue architecture by immunohistochemistry. Researchers should optimize working dilutions empirically based on sample preparation and detection chemistry.
