The MAPK/ERK signaling cascade controls proliferation, differentiation, and survival responses in many cell types, and MEK1 (MAP2K1) sits at the center of that pathway, phosphorylating ERK1/2 downstream of Ras and Raf. Threonine 286 is an autoregulatory phosphorylation site linked to MEK1 catalytic activity, so phospho-specific antibodies give a direct read on pathway activation in cell lysates and tissue extracts.

This unconjugated rabbit polyclonal reagent from Antibodies.com was raised against a phosphorylated threonine 286 epitope on MEK1. It is supplied at 1 mg/mL in PBS with 0.1% sodium azide as preservative and 50% glycerol as cryoprotectant, and ships in a 100 µL vial. The antibody has been validated for Western blotting and shows reactivity with human, mouse, and rat samples, supporting work across established cell lines, primary cultures, and rodent tissue models commonly used in signaling research.

Store the vial at -20 °C in working aliquots to limit freeze-thaw cycles and protect the phospho-epitope from inadvertent dephosphorylation during handling. For western blot, dilute into standard BSA or non-fat milk blocking buffer and probe PVDF or nitrocellulose membranes carrying SDS-PAGE-resolved lysates; a goat anti-rabbit HRP-conjugated secondary will detect the unconjugated primary. The polyclonal format yields recognition of multiple epitopes around the phospho-T286 region, which can improve signal when nearby sequence variation exists between species or when the modification state shifts dynamically in response to serum, growth factor, or pharmacological stimulation of the MAPK module.
