Raised in rabbit against a synthetic phosphopeptide corresponding to the sequence surrounding threonine 319 of human MEF2A (V-T-T(p)-P-S), this polyclonal IgG reagent from Antibodies.com isolates the transcriptionally active, phosphorylated form of MEF2A. Affinity chromatography against the phospho-T319 epitope enriches specific binders, while a subtractive pass over the non-phospho peptide removes cross-reactive antibodies that recognize unmodified MEF2A, giving clean detection of the Thr319-modified pool in immunofluorescence experiments. The reagent is supplied at 1.0 mg/mL in PBS pH 7.4 with 150 mM NaCl, 0.02% sodium azide, and 50% glycerol, and is packaged as a 50 µl vial. Phosphorylation at Thr319 (and the adjacent Thr312) is deposited by MAPK14/p38α and MAPK11/p38β in response to stress, growth factors, and hypertrophic cues, and is the modification that switches MEF2A from a HDAC-repressed state into an active driver of muscle and neuronal gene programs. Researchers following this activation event in cultured Human, Mouse, or Rat cardiomyocytes, myotubes, or fixed brain sections can therefore use the readout to track CaMK and p38 MAPK pathway engagement at the level of MEF2A T319 phosphorylation. The unconjugated primary antibody is shipped at 4 °C; on arrival, aliquot and store at -20 °C, protect from light, and limit freeze/thaw cycles to preserve binding activity across downstream imaging runs.
