This rabbit polyclonal antibody from Antibodies.com specifically recognizes MDM2 only when phosphorylated at serine 166, a modification that drives MDM2 nuclear import and strengthens its E3 ubiquitin ligase activity toward p53. The reagent is raised in rabbit and supplied unconjugated at 1 mg/ml in phosphate buffered saline (PBS) containing 0.05% sodium azide at approximately pH 7.2. It is validated for Western blot (WB) and immunohistochemistry (IHC) workflows and shows reactivity with human, mouse, and rat samples.

Phosphorylation at S166 is carried out primarily by AKT/PKB downstream of PI3K signaling, so phospho-S166 MDM2 serves as a useful readout for growth factor and insulin pathway activation, DNA damage-associated crosstalk, and oncogenic signaling in tumor biology. Laboratories studying the p53-MDM2 negative feedback loop, chemotherapeutic sensitization, or regulation of this tumor suppressor use the antibody to monitor pathway activation in cell lysates by WB and to localize phosphorylated MDM2 in tumor and normal tissue sections by IHC. The polyclonal format increases the chance of detecting the modified epitope across a range of surrounding sequence contexts.

The 100 µl pack size supports multiple blotting membranes or dozens of stained slides, and the azide-containing buffer can be applied directly in standard immunohistochemistry protocols without further manipulation of the preservative component.
