Anti-MDM2 (phospho S166) Antibody

Raised in rabbit against a synthetic phosphopeptide corresponding to the region of human MDM2 surrounding serine 166, this unconjugated polyclonal IgG recognizes endogenous MDM2 only when that residue is phosphorylated, leaving the non-phosphorylated form undetected. Affinity purification against the immunizing phosphopeptide enriches the phospho-specific fraction, supporting clean readouts of activated MDM2 within the p53–MDM2 regulatory axis. Cross-reactivity is documented for human, mouse and rat samples, so the same reagent can be used to compare DNA damage responses, oncogenic stress signaling or kinase inhibitor treatment across cell lines and rodent tissue without switching primaries. The reagent is supplied as a 200 µl liquid aliquot in phosphate-buffered saline (pH 7.4) with 150 mM NaCl, 0.02% sodium azide and 50% glycerol, shipped at 4 °C. Upon arrival it should be aliquoted and stored at -20 °C, with freeze/thaw cycles avoided to preserve phospho-epitope binding. In western blot, the antibody resolves the characteristic MDM2 band, and pairing it with a total-MDM2 reagent and an appropriate loading control allows relative S166 phosphorylation to be quantified. In immunofluorescence on fixed and permeabilized cells, the same antibody reveals the subcellular distribution of activated MDM2, and working dilution should be titrated empirically for each sample and detection chemistry.
