MCM7 (Minichromosome Maintenance Complex Component 7) is one of six core subunits of the MCM2-7 hexameric replicative helicase that loads onto eukaryotic replication origins and licenses each origin for a single round of DNA synthesis per cell cycle. This rabbit polyclonal preparation from Antibodies.com recognizes the MCM7 protein and is qualified for western blot, immunohistochemistry, and immunofluorescence detection of the endogenous target in human, mouse, and rat specimens.
The reagent ships as 100 µl of liquid at 1 mg/ml formulated in PBS with 0.1% sodium azide as a microbial preservative and 50% glycerol as a cryoprotectant, and should be stored at -20°C between uses. Aliquot upon first thaw to minimize repeated freeze-thaw exposure; the polyclonal nature of the antibody means it engages multiple independent epitopes, which can dampen the impact of any single conformational change caused by handling. Sodium azide is present for routine bench-top convenience, and the unconjugated format lets the vial be paired downstream with any HRP, biotin, or fluorophore-labeled secondary of the user's choice.
On western blot of whole-cell lysates, the antibody detects an endogenous band at the expected mass for MCM7 and tolerates standard RIPA or SDS sample buffer conditions. In immunohistochemistry on formalin-fixed paraffin-embedded tissue, staining concentrates in nuclei of proliferating compartments, matching the chromatin-associated function of MCM2-7 during replication licensing. In immunofluorescence on cultured cells, the same antibody produces a diffuse nuclear signal that intensifies through S phase in synchronized populations. Because MCM7 expression tracks proliferative activity and is extensively deployed as a sensitive marker in tumor biology studies, broad reactivity with mouse and rat samples in addition to human supports orthologous comparisons of replication licensing and origin firing across model systems.
