The Anti-MARCKS (phospho-S163) Antibody from Antibodies.com is a rabbit polyclonal reagent raised against the myristoylated alanine-rich C-kinase substrate (MARCKS) phosphorylated at serine 163, a key regulatory residue modified by protein kinase C (PKC) in cells and tissues. MARCKS is one of the most prominent cellular PKC substrates: in its non-phosphorylated state it sequesters calmodulin and tethers actin filaments to the inner leaflet of the plasma membrane, and phosphorylation at S163 (and the nearby S152, S156, and S170 residues) releases it from the membrane, making phospho-S163 MARCKS a widely used readout of acute PKC pathway activity in cell-signaling research.

Supplied at 1 mg/ml in PBS with 0.1% sodium azide and 50% glycerol, the antibody is unconjugated and validated for Western blot (WB) and immunohistochemistry (IHC) applications, with reactivity confirmed against human, mouse, and rat samples. This cross-species recognition allows consistent phospho-epitope detection across rodent disease models and human clinical specimens within the same laboratory workflow. The polyclonal format increases the probability of robust binding to the phospho-S163 epitope even when flanking residues differ between species.

Researchers use this reagent to study dynamic MARCKS phosphorylation in long-term potentiation and hippocampal synaptic plasticity, growth-cone guidance during neuronal development, macrophage and microglial activation, smooth muscle contraction, and regulated mucus secretion from airway and intestinal epithelia. Each vial contains 100 µl of antibody, sufficient for multiple rounds of membrane probing and slide staining. For Western blot, lysates should be prepared with freshly added phosphatase inhibitors to preserve the labile phospho-S163 modification before SDS-PAGE separation and transfer to PVDF or nitrocellulose membranes.
