The phospho-specific antibody from Antibodies.com targets the dual-phosphorylated activation loop of the JNK family of stress-activated kinases, recognizing MAPK8 (JNK1), MAPK9 (JNK2), and MAPK10 (JNK3) at residues T183/Y185 and T221/Y223 within the conserved Thr-X-Tyr motif. Raised in rabbit as a polyclonal IgG and supplied unconjugated at 50 µl per vial, this reagent is validated for western blot and immunofluorescence across human, mouse, and rat samples.

Because the antibody selectively binds the activated, phosphorylated form of JNK, it enables researchers to monitor c-Jun N-terminal kinase signaling downstream of cellular stress, inflammatory cytokines, and growth factor cues. The T183/Y185 and T221/Y223 sites are phosphorylated by the upstream MKK4 and MKK7 kinases, so detection of phospho-JNK provides a direct readout of pathway activation status rather than merely total JNK expression levels. This dual-phosphorylation specificity is useful for tracking stimulus-dependent signaling kinetics in cultured cells and tissue sections.

For best results, aliquot the antibody upon first use and store at -20 °C to limit repeated freeze-thaw cycles and preserve reagent performance. Optimal working dilutions should be determined empirically for each sample type, whether applied to western blot membranes or immunofluorescence preparations.
