This polyclonal rabbit IgG antibody from Antibodies.com detects the dually phosphorylated, activated forms of the JNK family of stress-activated protein kinases. It recognizes MAPK8 (JNK1) and MAPK9 (JNK2) only when phosphorylated at the activation-loop residues threonine 183 and tyrosine 185, and MAPK10 (JNK3) when phosphorylated at the corresponding threonine 221 and tyrosine 223 sites. Because activation requires phosphorylation of both the threonine and tyrosine within the TPY motif, the reagent specifically reports on catalytically engaged kinase, not total JNK expression.

The unconjugated antibody is supplied as a 100 µl volume and is validated for Western blot (WB) and immunofluorescence (IF) applications. Reported reactivity spans human, mouse, and rat samples, making it suitable for comparative work across cultured mammalian cells and tissue sections from these species. As a polyclonal reagent, it offers dual coverage of the JNK1 and JNK2 phospho-epitopes within a single preparation, which is useful when probing total stress-activated JNK signaling without the need to distinguish individual isoforms.

Typical use cases include tracking JNK pathway activation following cellular insults such as UV irradiation, oxidative stress, cytokine stimulation, or osmotic shock, as well as characterizing kinase cascade engagement in signaling studies and neuroscience research, where the JNK3 isoform is of particular interest. Aliquot upon first use and store refrigerated to preserve phospho-specific binding activity; avoid repeated freeze-thaw cycles, which can degrade recognition of these labile phospho-epitopes. For research use only.
