Raised in rabbit against a synthetic phosphopeptide derived from human p38/MAPK14 centred on the threonine 180 residue, this unconjugated polyclonal IgG reagent selectively recognises the activated pool of MAPK14 (p38α) when it has been phosphorylated on Thr180 within the kinase activation loop. Because the epitope is phosphorylation-dependent, the antibody discriminates phospho-p38α from total p38α protein in cell lysates and tissue samples, supporting studies of cellular stress, inflammatory cytokine, and UV responses where MKK3/MKK6-driven Thr180 phosphorylation is being measured.

Supplied by Antibodies.com as a 100 µl liquid aliquot at approximately 1 mg/ml in PBS with 0.1% sodium azide and 50% glycerol, the reagent is validated for western blot and immunofluorescence workflows. The target band migrates at approximately 42 kDa, consistent with full-length MAPK14. The antibody has been affinity-purified by epitope-specific chromatography to a purity of 95% by SDS-PAGE and is reported to detect the phosphorylated kinase in samples of both human and rat origin.

For western blot, lysates should be prepared in the presence of phosphatase inhibitors to preserve the labile Thr180 phosphate, and untreated versus stimulated samples (for example, cells exposed to TNF-α, IL-1β, or anisomycin) are typically run side-by-side as negative and positive controls. The unconjugated format is compatible with standard anti-rabbit IgG-HRP secondary reagents for chemiluminescent detection and with anti-rabbit IgG-fluorophore conjugates for immunofluorescence imaging. The reagent is intended for research use only and should be stored refrigerated; avoid repeated freeze-thaw cycles of the glycerol-containing aliquot to maintain antibody activity.
