This polyclonal antibody from Antibodies.com targets microtubule-associated protein 2 (MAP2), a high-molecular-weight cytoskeletal protein enriched in neuronal dendrites and widely used as a marker of post-mitotic neuron identity in neuroscience research. Raised in rabbit and supplied as an unconjugated IgG preparation in a 100 µl volume, the reagent is validated for Western blot detection of MAP2 in human and mouse sample lysates, including brain and spinal cord tissue, primary neuronal cultures, and differentiated neuron-like cell lines. MAP2 stabilizes microtubules in the dendritic compartment, so the antibody is frequently applied to studies of neuronal differentiation, axonal versus dendritic specification, cytoskeletal remodeling after injury, and characterization of neural stem cell-derived neurons. The polyclonal format provides multiple epitope recognition, which can be useful when probing conserved regions of MAP2 isoforms (MAP2a, MAP2b, MAP2c, MAP2d) across species. For Western blot, samples are typically homogenized in RIPA or similar lysis buffer, separated by SDS-PAGE, and transferred to nitrocellulose or PVDF membranes before blocking and probing; expected band sizes for the high-molecular-weight MAP2 isoforms are generally observed in the 280 kDa region of the gel. Researchers are advised to titrate the antibody across a range of dilutions to optimize signal-to-noise for their particular sample type and transfer conditions.
