A rabbit polyclonal IgG reagent from Antibodies.com raised against MAP1LC3B, the microtubule-associated protein 1 light chain 3 beta that serves as the canonical autophagosome marker in mammalian cells. Supplied as a 200 µL unconjugated liquid aliquot, this primary antibody is validated for western blot (WB) and immunofluorescence (IF) workflows on human, mouse, and rat samples, giving laboratories a single reagent that spans the most common cell and animal models used in autophagy research.

The polyclonal antibody detects both isoforms of LC3B generated during autophagosome biogenesis: the cytosolic precursor LC3B-I (~17 kDa) and the lipidated, autophagosome-associated LC3B-II (~14 kDa). Resolving the two bands together on a western blot membrane, or scoring discrete LC3B puncta in fixed cells by immunofluorescence, supports flux measurements that distinguish autophagosome formation from clearance. The polyclonal format amplifies signal on the smaller LC3B-II fragment and enables robust detection of endogenous expression in cell and tissue lysates without the need for overexpression.

For storage, each vial should be aliquoted and kept at -20 °C to protect against repeated freeze-thaw cycles; working aliquots are refrigerated short-term and brought back to room temperature before use. In western blot, primary incubation at room temperature with a standard HRP-conjugated Goat Anti-Rabbit IgG secondary and ECL chemiluminescence is typical on standard SDS-PAGE gels loaded at roughly 25 µg of total protein per lane; in immunofluorescence, counterstaining nuclei with DAPI aids visualisation of LC3B puncta relative to subcellular structures. Combining the antibody with lysosomal inhibitors such as bafilomycin A1 or chloroquine confirms autophagic flux by demonstrating further LC3B-II accumulation, and pairing it on the same blot with p62/SQSTM1 or PARP1 antibodies builds a fuller picture of the autophagy pathway. The product is supplied for research use only.
