Raised in rabbit against the autophagy marker MAP1LC3A (microtubule-associated protein 1 light chain 3 alpha), this unconjugated IgG reagent from Antibodies.com is a recombinant monoclonal antibody (#A11428) that detects both the cytosolic LC3-I precursor (~18 kDa) and the phosphatidylethanolamine-conjugated, autophagosome-associated LC3-II form (~14-16 kDa). The reagent is supplied as 200 µl of liquid in a vial formulated in PBS with glycerol and sodium azide, and should be stored at -20°C with aliquoting recommended to limit freeze-thaw cycles.

Validated for western blot, immunohistochemistry, immunofluorescence, and immunoprecipitation on human, mouse, and rat samples, the antibody tracks autophagic flux through the LC3-I to LC3-II conversion catalyzed by the E1-like Atg7 and E2-like Atg3 enzymes. In immunofluorescence imaging, it produces the characteristic punctate cytoplasmic staining that marks autophagosomal membranes, with applications in neurodegeneration, cancer cell biology, and metabolic stress models.

For research use only. Pair with bafilomycin A1 or chloroquine treatment when measuring flux, since these lysosomal inhibitors cause LC3-II accumulation that can be quantified by western blot band intensity.
