The A11428 clone from Antibodies.com is a rabbit monoclonal IgG antibody raised against MAP1LC3A (also known as LC3A), one of three human ATG8-family homologs (LC3A, LC3B and LC3C) routinely monitored as a marker of autophagosome formation. Supplied as 100 µL of unconjugated antibody, the reagent is validated for western blot, immunohistochemistry, immunofluorescence, and immunoprecipitation workflows on human, mouse and rat samples.

MAP1LC3A is cleaved at its C-terminus by ATG4 proteases to generate the cytosolic LC3A-I form, then conjugated to phosphatidylethanolamine during autophagy induction to produce the membrane-associated LC3A-II species that decorates autophagosomal membranes. Tracking the LC3A-II band shift and the appearance of punctate LC3A staining are standard readouts in starvation, mTOR-inhibition and mitophagy studies, as well as in screens for compounds that perturb autophagic flux.

The unconjugated format leaves the choice of reporter to the user, so the same vial can be paired with HRP- or fluorophore-labeled secondary antibodies for chromogenic western blot development, immunofluorescence imaging of cytoplasmic puncta, antigen retrieval-based IHC on fixed tissue sections, or IP of endogenous LC3A complexes prior to mass spectrometry and co-immunoprecipitation analyses. Host species is rabbit, isotype IgG, supplied as a liquid formulation at 100 µL per vial.
