Mitotic Arrest Deficient 2 (MAD2) sits at the heart of the spindle assembly checkpoint (SAC), where it monitors kinetochore-microtubule attachment and delays anaphase onset until every chromosome has achieved proper bipolar attachment. MAD2 cycles between open and closed conformations and, together with BUBR1, BUB3, and CDC20, assembles the mitotic checkpoint complex (MCC) that inhibits the anaphase-promoting complex (APC/C). Loss or dysregulation of MAD2 is linked to chromosomal instability and aneuploidy in many tumor types, making it a widely studied marker in cell cycle and cancer research.

From Antibodies.com, this unconjugated rabbit polyclonal primary antibody is supplied as a 100 µl liquid aliquot and is validated for Western blot and ELISA workflows. It was raised against human MAD2 and is intended for research use only. Each lot is affinity-purified to minimize non-specific background, and the antibody performs cleanly on immunoblots of cell lysates prepared from proliferating human cell lines such as HeLa or HEK293. For long-term storage, aliquot upon receipt and keep working stocks at -20 °C; limit freeze-thaw cycles to preserve activity.

Typical applications include probing MAD2 expression in cells arrested in mitosis with nocodazole or Taxol, comparing checkpoint-proficient versus checkpoint-deficient populations, and tracking MCC assembly in immunoprecipitation experiments alongside anti-CDC20 and anti-BUBR1 antibodies. The reagent is also useful for monitoring SAC integrity, mitotic slippage, and chromosome missegregation phenotypes in both lysate-based and fixed-cell assay formats.
