Validated for Western blot and immunohistochemistry work, this unconjugated rabbit polyclonal antibody from Antibodies.com targets the Lyn tyrosine kinase and detects the protein in human, mouse, and rat samples. Lyn is a non-receptor Src-family kinase (approximately 56 kDa) that mediates both positive and negative signaling downstream of the B-cell receptor, FcεRI in mast cells, and several cytokine receptors, making it a frequent target in studies of hematopoietic signaling, autoimmune disease, and lymphoma biology.

The reagent is supplied at 1 mg/ml in PBS with 0.1% sodium azide and 50% glycerol, in a 100 µl vial. The cryoprotectant buffer supports storage at 4 degrees C for short-term working aliquots or at -20 degrees C for long-term stability, kept protected from light and limited to freeze-thaw cycles by aliquoting on receipt. The polyclonal format allows the reagent to recognize multiple epitopes across the kinase region, which is helpful when probing truncated isoforms or denatured antigens in fixed tissue sections prepared for IHC. For Western blot applications, a typical workflow involves lysing cells in RIPA buffer, resolving 20-40 µg of total protein by SDS-PAGE, transferring to nitrocellulose or PVDF, and probing at empirically determined dilutions followed by an HRP- or fluorophore-conjugated secondary.

Research applications include tracking Lyn phosphorylation kinetics downstream of FcεRI engagement in mast cells, characterizing B-cell signaling defects in Lyn-deficient mouse models, and profiling Lyn expression across lymphoid and myeloid tissue by immunohistochemistry. Each lot is produced by peptide immunization of rabbits and peptide-affinity purification to reduce off-target reactivity. This primary antibody is intended for research use only and is not validated for clinical diagnostic or therapeutic applications.
