Anti-LRRK2 Antibody

Raised in rabbit as a polyclonal IgG, this unconjugated primary antibody from Antibodies.com recognizes human leucine-rich repeat kinase 2 (LRRK2), a ~286 kDa multidomain kinase encoded on chromosome 12 (PARK8) and most strongly implicated in autosomal-dominant Parkinson disease. The reagent is supplied as 100 µl of liquid, validated for western blot (WB), immunohistochemistry (IHC), and flow cytometry (FC) workflows.

LRRK2 contains an N-terminal armadillo and ankyrin repeat region, a central leucine-rich repeat (LRR) domain, and a C-terminal ROC–COR–kinase–WD40 catalytic core that phosphorylates a subset of Rab GTPases governing membrane trafficking. The kinase-domain G2019S variant, which is responsible for roughly 80% of LRRK2-linked PD cases and increases kinase activity about two-fold, and the ROC-domain R1441C/G/H substitutions, which activate the enzyme roughly three- to four-fold, are commonly probed with this reagent.

Investigators apply this LRRK2 antibody to track endogenous protein expression in iPSC-derived dopaminergic neurons and patient-derived neutrophils, characterize microglial and inflammatory signaling in neurodegeneration models, map α-synuclein crosstalk, and follow Ser1292 autophosphorylation as a pharmacodynamic biomarker of LRRK2 inhibitor treatment.
