Raised in rabbit against a fusion protein corresponding to internal residues of human leprecan-like 2, this polyclonal IgG reagent from Antibodies.com recognises endogenous LEPREL2, a prolyl 3-hydroxylase family member (also called Prolyl 3-hydroxylase 3 and Leprecan-like protein 2) that participates in collagen post-translational modification within the endoplasmic reticulum. The antibody is antigen-affinity-purified and supplied unconjugated in a 200 µl vial at approximately 2.2 mg/ml in PBS (pH 7.3) with 50% glycerol and 0.05% sodium azide, leaving secondary-reagent choice (HRP, fluorophore, biotin) to the end user. Validation data on human A549 and HeLa cells and on mouse heart tissue confirm reactivity with human and mouse samples in western blot, with representative westerns run on 6% SDS-PAGE using 40 µg of lysate per lane at a 1:300 primary dilution and an 8000-fold diluted secondary for roughly 20 seconds of exposure; the unconjugated IgG format also supports downstream chemiluminescent or fluorescent detection on PVDF or nitrocellulose membranes. Recommended handling is to aliquot upon first thaw to limit freeze-thaw cycles, store refrigerated between uses, protect from prolonged light exposure, and hold long term at -20 °C in the glycerol-containing formulation. Applications include studies of basement-membrane collagen biosynthesis, hydroxylation-defect phenotypes in connective tissue, and comparative expression across the LEPREL gene family in cell lysates and tissue extracts.
