The phospho-Y255 epitope of LAT (Linker for Activation of T cells) is one of the earliest ZAP-70 substrates modified after T-cell receptor (TCR) engagement, and tracking this specific phospho-site is a standard way to monitor early T-cell activation. This unconjugated rabbit polyclonal antibody from Antibodies.com is raised against that phospho-site and is validated for Western blot detection in human, mouse, and rat samples.

LAT is a 36–38 kDa transmembrane adapter that, upon TCR triggering, becomes phosphorylated on multiple tyrosines by ZAP-70. Y255 in particular sits within a cluster of distal tyrosines that recruit SH2-domain effectors, propagate calcium signaling, and feed the Ras/MAPK and NFAT pathways downstream of the immune synapse. Detecting the Y255 phospho-form is therefore useful for comparing TCR signaling strength between wild-type and knockout T-cell lines, profiling primary lymphocyte activation, and benchmarking inhibitors of the Src/ZAP-70 kinase cascade.

The reagent is supplied in a vial as 100 µL of a 1 mg/ml stock in phosphate-buffered saline (PBS) at approximately pH 7.2, with 0.05% sodium azide as a preservative. For Western blot, phospho-enriched or stimulated lysates give stronger signal than resting samples. Short-term storage in the refrigerator is fine; for longer-term use, aliquot the antibody and freeze at -20°C to limit freeze-thaw cycles, and protect from prolonged light exposure. Intended for research use only.
