Ribosomal-targeting cytokeratin KRT16 is a type I intermediate filament protein expressed in the suprabasal layers of stratified squamous epithelia, where it is widely used as a readout of keratinocyte hyperproliferation, wound re-epithelialization, and abnormal epidermal differentiation. The Antibodies.com anti-KRT16 polyclonal reagent is raised in rabbit (IgG) and supplied as 50 µl of unconjugated antiserum, making it compatible with both chromogenic and fluorescence detection chemistries. Validated applications include western blot (WB) for resolving KRT16 from cultured keratinocytes, organotypic skin equivalents, and epithelial lysates, and immunohistochemistry (IHC) on formalin-fixed tissue sections for mapping suprabasal keratinocyte compartments.

Species reactivity covers human, mouse, and rat, enabling translational comparison between patient biopsies and rodent models of skin disease. Common research uses include characterization of pachyonychia congenita and related keratin mutation disorders, profiling psoriatic or wounded skin, oral mucosal keratinization studies, and labelling of hyperproliferative keratinocytes in stratified epithelial tissue. The unconjugated, polyclonal format supports flexible secondary antibody pairing and allows affinity-based amplification of the KRT16 signal when antigen abundance is low.
