Anti-KPNA4 Antibody [3D10]

Beyond basic identification, clone 3D10 was raised against recombinant full-length mouse KPNA4 protein and, notably, shows no detectable cross-reactivity with the closely related paralogues importin 1/Rch1, importin 5/NPI-1, or importin 7/S2 when tested against GST fusion proteins (Sakaguchi N. et al., Hybrid. Hybridomics 22(6):397-400, 2003). The same study validated endogenous Qip1 detection by Western blot in lysates from human HeLa, monkey COS-7, mouse L-929, rat NRK, hamster BHK, and bovine MDBK cells, supporting the broad reactivity panel listed by Antibodies.com. For Western blot the manufacturer recommends a working dilution of 1:250-1:500 in standard Laemmli gels, and 10 µg of total cell lysate per lane is a typical loading amount. KPNA4 belongs to the importin-α subfamily 3 together with KPNA3 and acts as the principal adaptor that ferries the NF-κB p50/p65 heterodimer, p53, MeCP2 (relevant to Rett syndrome), and SV40 T antigen into the nucleus; it is also exploited by influenza A virus, which makes this antibody useful in cancer signalling, viral entry, and neurodevelopmental research. The reagent is supplied as a 200 µg aliquot in liquid format and should be refrigerated at 4 °C for short-term use or aliquoted and stored at -20 °C to limit freeze-thaw cycles.
