KNTC2 (also called KNL1) is a kinetochore scaffold protein that anchors the spindle assembly checkpoint machinery and is required for accurate chromosome segregation during mitosis. The rabbit polyclonal antibody from Antibodies.com was raised against the human KNTC2 sequence and cross-reacts with the mouse ortholog, supporting comparative work between human cell lines and murine tissues. Validated readouts include Western blot for protein-level analysis and immunofluorescence (IF) for visualizing kinetochore localization in fixed cells.

Each vial ships as 100 µL of unconjugated antiserum, leaving the choice of reporter (HRP, biotin, or a fluorophore) to the researcher. The unconjugated format suits both chemiluminescent WB workflows and high-resolution IF imaging of mitotic chromosomes. For storage, aliquot upon arrival and freeze at -20°C; working dilutions can be kept refrigerated short term at 4°C, with protection from light recommended when fluorescent secondary conjugates are used downstream. Limiting freeze-thaw cycles helps preserve antibody titer over many experiments.

KNTC2 acts as the central landing pad for BUB1-BUB3 recruitment through its MELT-repeat motifs, sits within the KMN network (KNL1-Mis12-Ndc80), and is therefore a routine marker in studies of chromosome instability, aneuploidy in cancer cells, mitotic checkpoint strength, and kinetochore-microtubule attachment. The endogenous protein migrates as a high-molecular-weight band on SDS-PAGE, consistent with its large scaffolding size in the 200-250 kDa range.
