Raised in rabbit against human kallikrein-related peptidase 5, this unconjugated polyclonal IgG reagent from Antibodies.com is supplied as a 200 µl aliquot and optimized for Western blot detection of the native antigen. KLK5 is a secreted trypsin-like serine protease best characterized in the epidermis, where it initiates a protease cascade that activates KLK7, KLK14, and related kallikreins to drive corneodesmosome shedding and orderly desquamation. Activity of KLK5 is normally restrained by the SPINK5-encoded inhibitor LEKTI; when this brake fails, unopposed KLK5 activity contributes to the stratum corneum barrier defect seen in Netherton syndrome, and elevated KLK5 levels have been reported in lesional tissue from atopic dermatitis and psoriasis patients. The protease is also expressed in esophageal epithelium, salivary gland, and several hormone-responsive tissues, and is studied in ovarian, breast, and prostate cancer research as a candidate biomarker of tumor microenvironment remodeling. The antibody is unconjugated and isotype IgG, leaving the choice of secondary reporter and detection chemistry to the user; for routine immunoblotting, a working dilution in the 1:500–1:2000 range is a reasonable starting point and should be titrated against the abundance of KLK5 in the preparation. Each vial is supplied at 200 µl; store the stock frozen at -20 °C in single-use aliquots to limit freeze-thaw cycles, and keep working solutions refrigerated during short-term use.
