Keratin 8 (K8) is a type II intermediate filament protein expressed predominantly in simple and glandular epithelia, where it heterodimerizes with K18 to form the cytoskeletal scaffolding of hepatocytes, intestinal lining cells, and most carcinoma cell lines of epithelial origin. This unconjugated polyclonal reagent from Antibodies.com was raised in rabbit and is validated for western blot (WB), immunohistochemistry (IHC), and immunofluorescence (IF) workflows on human samples. The antibody recognizes the K8 polypeptide at the Ab-74 epitope, allowing researchers to track total K8 levels in cell lysates, map intermediate filament architecture in fixed tissue, and visualize cytoskeletal reorganization in cultured epithelial cells.

K8 is widely used as a marker of epithelial differentiation and as a substrate for studying stress-activated kinases, since its N-terminal head domain carries several phosphorylation sites that remodel filaments during mitosis, oxidative stress, and apoptosis. Laboratories working on hepatocellular biology, intestinal epithelial barrier function, and epithelial-derived carcinomas commonly pair this reagent with K18 antibodies to assess epithelial integrity, or with phospho-specific reagents to dissect signaling cascades that converge on intermediate filaments. The unconjugated format allows end users to couple the antibody to HRP, biotin, Alexa Fluor dyes, or other reporter systems depending on assay design.

Supplied as a 100 µL liquid vial, the reagent is stored under standard short-term antibody handling conditions and should be aliquoted to limit freeze-thaw cycles across multiple experiments. Reactivity is confirmed for human K8.
