This phospho-specific polyclonal reagent targets the dual activation-loop phosphorylation motif (threonine 183 and tyrosine 185) shared by JNK1, JNK2, and JNK3 (MAPK8/9/10). The Antibodies.com antibody selectively recognizes the activated, dually phosphorylated kinase and does not detect the inactive unphosphorylated form, making it a useful reagent for monitoring JNK pathway engagement in response to cellular stress, inflammatory cytokines, UV irradiation, or chemotherapeutic challenge.

Validated for Western blot (WB), the antibody is supplied at 1 mg/ml in phosphate buffered saline (PBS) containing 0.05% sodium azide at approximately pH 7.2. The 100 µL pack size supports multiple membrane probing experiments. Recommended starting dilutions generally range from 1:500 to 1:2000 depending on sample abundance and detection chemistry (chemiluminescence or near-infrared). To preserve the phospho-epitope, lysates should be prepared in the presence of phosphatase inhibitors and used promptly or stored frozen; lambda phosphatase treatment of samples should be avoided because it removes the target modification.

Raised in rabbit, the reagent has been tested for cross-reactivity with human, mouse, and rat samples, covering the principal model systems used in stress-kinase research. JNK family kinases phosphorylate c-Jun and other transcription factors, regulating apoptosis, proliferation, and inflammatory gene expression. Investigators studying neurodegeneration, cancer cell signaling, insulin resistance, or cardiac stress pathways frequently employ phospho-JNK antibodies to measure pathway activation status alongside total JNK levels.
