Generated in rabbit against a synthetic peptide corresponding to a region surrounding V391 of human IRF-3, this polyclonal antibody from Antibodies.com is intended for detection of endogenous Interferon Regulatory Factor 3 in Western blot and immunohistochemistry workflows.

IRF-3 is a central transcription factor of the innate antiviral response. On detection of viral RNA by RIG-I-like receptors or activation of the cGAS-STING DNA-sensing pathway, IRF-3 is phosphorylated by TBK1, dimerizes, and translocates to the nucleus where it drives type I interferon and interferon-stimulated gene expression. Anti-IRF-3 reagents are widely used to monitor this activation step in infected cells, primary immune cell populations, and relevant tissue sections.

The antibody was raised against a human IRF-3 peptide and cross-reacts with mouse and rat IRF-3, supporting comparative innate immunity studies across these species. It is supplied unconjugated, leaving the immunoglobulin available for downstream HRP or fluorophore conjugation or direct use with chromogenic and fluorescence-based detection chemistries.

The product is provided at 1 mg/ml in phosphate buffered saline containing 0.05% sodium azide at approximately pH 7.2, in a 50 µl vial sized for typical research use. Working aliquots are stable when refrigerated; the azide additive guards against microbial contamination. For Western blot, samples are commonly prepared in standard RIPA or NP-40 lysis buffer supplemented with protease and phosphatase inhibitors to preserve IRF-3 phosphorylation status, and for IHC the antibody is applied to formalin-fixed paraffin-embedded sections after heat-mediated antigen retrieval.
