This rabbit polyclonal reagent from Antibodies.com specifically recognizes the phosphorylated form of IKKα and IKKβ at serine 176, a site located within the activation loop of these kinases whose modification is required for full IKK complex activity and downstream canonical NF-κB signaling. Because the antibody was raised against a phospho-peptide, it discriminates activated IKK from the unphosphorylated pool in fixed tissue samples.

Supplied as 100 µl at 1 mg/ml in phosphate buffered saline (PBS) containing 0.05% sodium azide at approximately pH 7.2, the unconjugated antibody is validated for immunohistochemistry on human, mouse, and rat tissue sections. It can be paired with HRP-, biotin-, or fluorophore-conjugated secondary reagents for chromogenic or fluorescence detection in formalin-fixed, paraffin-embedded specimens or cryosections.

Phospho-epitopes are labile, so the vial should be stored refrigerated and protected from light; aliquot upon first thaw to limit repeated freeze-thaw exposure and preserve binding affinity for the phospho-S176 modification. The polyclonal format increases tolerance for minor sequence variation around the target residue, but researchers should still confirm staining with appropriate positive controls (for example, TNF-α-stimulated cells) alongside peptide-block or isotype controls. The product is intended for research use only.
