Raised in rabbit and affinity-purified, this unconjugated polyclonal reagent specifically recognizes the activated, phosphorylated form of IKKα and IKKβ at the activation-loop residues S176/177, the canonical phosphorylation sites within the IKK complex that drives canonical NF-κB signaling. Because detection depends on the phospho-epitope, the antibody supports signaling studies that follow IKK activation downstream of TNF-α, IL-1, LPS, and other pro-inflammatory stimuli, and complements total IKK blots by distinguishing the inactive kinase from its activated pool.

Validated for Western blot and immunohistochemistry, the antibody reacts with human and mouse samples, making it useful for cell-line work, primary-cell lysates, and archived tissue sections. The 100 µl pack is supplied at 1 mg/ml in phosphate buffered saline (pH ~7.2) containing 0.05% sodium azide as a preservative. Lysates should be prepared with phosphatase inhibitors to preserve the phospho-epitope, and the reagent should be stored refrigerated and protected from light, avoiding repeated freeze-thaw cycles. Sodium azide should be removed by dialysis or gel filtration before any conjugation step that uses amine-reactive chemistries.

Distributed by Antibodies.com, this reagent is suited to NF-κB pathway research, inflammation models, oncology investigations of IKK-driven survival signaling, and immune-cell activation studies where direct measurement of kinase activation is required.
