The phospho-specific polyclonal reagent from Antibodies.com targets the tyrosine 512 phosphorylation site on human ICAM1 (Intercellular Adhesion Molecule 1), a transmembrane immunoglobulin-family glycoprotein best known for mediating leukocyte adhesion to vascular endothelium. Raised in rabbit, the unconjugated IgG preparation is supplied at 100 µl and is validated for Western blot detection of the activated, Y512-phosphorylated form of ICAM1 in cell lysates.

Y512 sits within the cytoplasmic tail of ICAM1 and becomes phosphorylated downstream of Src-family kinases in response to inflammatory cues, oxidative stress, or mechanical stimulation of endothelial cells. Tracking this modification is therefore a way to monitor ICAM1 activation state rather than total expression, which is helpful when separating baseline adhesion-molecule abundance from signal-driven changes in endothelial or epithelial model systems.

Recommended use begins with standard SDS-PAGE separation of whole-cell lysates prepared in the presence of phosphatase inhibitors, followed by transfer to nitrocellulose or PVDF, blocking, and probing with the antibody at a working dilution appropriate for chemiluminescent detection. A total ICAM1 antibody can be run in parallel as a loading control to compare phosphorylated and total pools. The reagent is intended for research use only and should be stored refrigerated between uses to preserve phospho-specificity; avoid repeated freeze-thaw cycles by aliquoting on first thaw.
