This mouse monoclonal antibody from Antibodies.com targets the C-terminal cytoplasmic region of human ICAM-1 (Intercellular Adhesion Molecule 1, CD54), a glycosylated type I transmembrane glycoprotein expressed on endothelial cells, leukocytes, and activated epithelial cells. ICAM-1 mediates cell-cell adhesion by engaging the β2 integrins LFA-1 (CD11a/CD18) and Mac-1 (CD11b/CD18) on circulating leukocytes, and its ~90 kDa band on SDS-PAGE reflects heavy N-linked glycosylation of the extracellular Ig-like domains.

Validated for Western blot and immunocytochemistry, the unconjugated reagent is supplied as 100 µl of purified immunoglobulin in PBS at pH 7.4, supplemented with 0.02% sodium azide as a preservative and 50% glycerol for cryoprotection. It reacts specifically with human ICAM-1 and detects both full-length protein and shorter C-terminal fragments generated by shedding or proteolytic cleavage in cell lysates and fixed-cell preparations.

ICAM-1 transcription is strongly induced by pro-inflammatory cytokines such as TNF-α, IL-1β, and IFN-γ, making the protein a widely used marker of endothelial activation. Investigators use C-terminal-specific antibodies to study leukocyte transmigration across vascular monolayers, neutrophil and lymphocyte recruitment in inflamed tissue, and signal transduction events downstream of ICAM-1 clustering, including Src-family kinase activation, Rho GTPase signaling, and cytoskeletal reorganization. Reported applications include models of atherosclerosis, allograft rejection, sepsis, and chronic inflammatory disease, where changes in ICAM-1 abundance and cleavage are tracked in cultured endothelial and epithelial cells.
