HSF1 (Heat Shock Factor 1) is the master transcriptional regulator of the cellular heat shock response, controlling expression of HSP70, HSP90, and other molecular chaperones that maintain proteostasis under thermal, oxidative, and proteotoxic stress. This Antibodies.com reagent is a rabbit polyclonal antibody raised against a region of human HSF1 corresponding to residue 303, supplied as 100 µl of unconjugated antiserum suitable for Western blot and immunohistochemistry workflows on human samples.
The antibody recognizes an epitope within the regulatory domain of HSF1, a region that contains several mapped phosphorylation sites involved in modulating transcriptional activity, nuclear-cytoplasmic shuttling, and stress-dependent turnover. Researchers studying stress granule assembly, chaperone network dynamics, cancer cell survival pathways, and age-related protein aggregation use anti-HSF1 antibodies to monitor HSF1 levels and its post-translational regulation in cultured cell lines and primary tissues.
In Western blot, the reagent detects endogenous HSF1 migrating near its expected molecular weight of approximately 57 kDa on whole-cell lysates. For immunohistochemistry on formalin-fixed paraffin-embedded sections, antigen retrieval followed by incubation with this unconjugated primary supports nuclear staining consistent with HSF1's role as a DNA-binding transcription factor. Host species is rabbit, clonality is polyclonal, the conjugate is unconjugated, and the indicated reactivity is human; non-human samples should be tested empirically before use.
The product is supplied as a liquid in a 100 µl vial. For storage, keep the antibody frozen at -20°C and divide into single-use aliquots to minimize repeated freeze-thaw cycles; thawed aliquots can be held briefly at 4°C. The unconjugated format provides flexibility for downstream coupling to HRP, biotin, fluorophores, or gold conjugates chosen by the end user.
