HNRNPU, also known as scaffold attachment factor A (SAF-A), is a ubiquitous nuclear matrix protein whose N-terminal SAP domain binds scaffold attachment region (SAR) DNA while a C-terminal RGG-rich region engages RNA. The Mouse monoclonal antibody from Antibodies.com (clone A9907) recognizes the native protein in human samples and is validated for Western blot detection of HNRNPU in whole-cell lysates and chromatin-enriched fractions.

SAF-A contributes to long non-coding RNA retention on chromatin, including tethering of XIST and FIRRE lncRNAs to the inactive X chromosome, and supports PRC1- and PRC2-mediated histone modifications such as H2AK119ub and H3K27me3 on the Xi. Detection of HNRNPU therefore serves as a marker of nuclear integrity and as a reference in studies of X-chromosome inactivation, chromatin architecture, the DNA damage response, and pre-mRNA splicing dynamics.

Supplied as 200 µl of unconjugated mouse IgG, the reagent can be paired with HRP- or fluorophore-conjugated secondary antibodies. For best results, aliquot the antibody upon arrival and freeze for long-term storage; keep working stocks refrigerated and protect from light to preserve activity. Intended for research use only.
