Raised in rabbit against human hepatocyte nuclear factor 1-alpha (HNF-1 alpha), this unconjugated polyclonal antibody is supplied by Antibodies.com as a 100 µl liquid aliquot intended for western blot (WB) and immunohistochemistry (IHC) workflows. HNF-1 alpha is a POU-homeodomain transcription factor encoded by the HNF1A gene that drives hepatocyte and pancreatic beta-cell gene expression, including regulators of glucose transport and coagulation factor synthesis. Heterozygous loss-of-function variants in HNF1A cause maturity-onset diabetes of the young type 3 (MODY3), and altered HNF-1 alpha levels have been reported in hepatocellular carcinoma and other solid tumours, making reliable detection of the protein valuable for diabetes research, hepatic biology studies, and oncology investigations.

The reagent is provided without a fluorophore or enzyme conjugate, giving laboratories flexibility to pair it with their own secondary detection systems, such as HRP-based chromogenic IHC on formalin-fixed paraffin-embedded tissue sections or chemiluminescent western blot detection of the approximately 80 kDa HNF-1 alpha band. According to the supplier specifications, validated applications are western blot analysis of human cell and tissue lysates and immunohistochemistry on human tissue sections, with reactivity confirmed for the human target. Brief centrifugation before opening the vial helps recover the full 100 µl volume, and the contents can be diluted directly into standard blocking buffer or antibody diluent without further reconstitution.

For routine handling, store the vial at -20 °C in working aliquots to limit freeze-thaw cycles and protect from light during long-term storage. Users commonly titrate the antibody empirically against their own positive-control tissue (such as human liver or pancreas) and pair it with an appropriate rabbit-specific secondary reagent. Detailed lot-specific quality-control data accompany each shipment from Antibodies.com.
