The Antibodies.com polyclonal antibody to HLA-F is a rabbit-derived IgG reagent raised against the human non-classical MHC class I heavy chain HLA-F. Supplied as 200 µl of unconjugated antisera, it is validated for western blot and flow cytometry workflows on human samples and is intended for research use only.

HLA-F is structurally related to classical class I molecules but differs in its expression pattern: it is retained intracellularly in most resting cells and translocates to the plasma membrane upon activation, frequently appearing as a β2-microglobulin-free heavy chain or as a component of peptide-loaded open conformers. This reagent has been used to characterize HLA-F surface expression on extravillous trophoblasts, where it is thought to engage activating receptors on maternal NK cells, and on multiple tumor cell lines where dysregulated HLA-F expression has been linked to immune evasion.

For western blot, the antibody is typically incubated on reduced cell lysates, where it detects the approximately 40 kDa HLA-F heavy chain. For flow cytometry, it is suitable for surface and intracellular staining of human peripheral blood mononuclear cells and cultured lines. The polyclonal format offers broader epitope coverage than monoclonal alternatives, which can be useful when probing conformationally variable forms of HLA-F.

As an unconjugated IgG preparation, the antibody can be paired with a fluorescent or enzymatic secondary reagent of the researcher's choice. Aliquot upon first use and store refrigerated at 4 °C to preserve activity across multiple experiments.
