Raised in rabbit as a polyclonal reagent, this unconjugated primary antibody targets histone H3 tri-methylated at lysine 9 (H3K9me3), a repressive chromatin modification enriched at pericentric heterochromatin, repetitive elements, and silenced gene loci. From Antibodies.com, the antibody is supplied as 100 µl of liquid at 1 mg/ml in PBS containing 0.1% sodium azide and 50% glycerol, a storage buffer that protects antibody activity during routine bench use and longer-term freezer storage.

Validated applications cover western blot, immunohistochemistry, immunofluorescence, immunoprecipitation, chromatin immunoprecipitation (ChIP), and ChIP-seq, making the reagent useful for mapping H3K9me3 distribution across promoters, heterochromatic regions, and developmentally regulated loci in human, mouse, and rat samples. The tri-methyl K9 mark is commonly used as a reference for transcriptional repression, heterochromatin boundaries, and epigenetic reprogramming studies in differentiation, stress response, and disease models.

As an unconjugated preparation, the antibody is flexible for downstream detection chemistries: it can be paired with HRP-conjugated, biotinylated, or fluorophore-labelled secondary reagents for chromogenic western blot, IHC, and immunofluorescence readouts, or captured directly on protein A or protein G beads for ChIP and ChIP-seq workflows. The polyclonal host response increases the chance of recognising the tri-methyl modification across different flanking peptide contexts around the modified lysine, which can be helpful when comparing H3K9me3 patterns between species or experimental conditions.
