From Antibodies.com, this rabbit polyclonal IgG antibody recognises histone H3 symmetrically dimethylated at arginine 2 (di methyl R2). It is unconjugated and has been validated for western blotting (WB), immunohistochemistry (IHC), immunofluorescence staining (ICC/IF), immunoprecipitation (IP), chromatin immunoprecipitation (ChIP) and ChIP-seq. Reactivity has been confirmed in human, mouse and rat samples.

The immunogen is a synthetic symmetric dimethylated peptide corresponding to the region around R2 of human histone H3 (NCBI accession NP_003520.1), with peptide sequence MARTK. The target runs at a molecular weight of 17 kDa. The antibody is affinity purified and supplied as a liquid in phosphate buffered saline, pH 7.3, containing 50% glycerol and 0.01% thiomersal; concentration is reported as lot specific. Recommended working dilutions are WB 1:500–1:1,000, IHC 1:50–1:200, ICC/IF 1:50–1:200, IP 1:50–1:200, ChIP 1:20–1:100 and ChIP-seq 1:20–1:100. Representative western blots are run with the antibody at 1:1,000 alongside a goat anti-rabbit IgG H&L HRP secondary at 1:10,000, with 25 µg lysate per lane, 3% non-fat dry milk in TBST as blocking buffer, ECL Basic Kit detection and a 60 s exposure. IHC validation is shown on paraffin-embedded rat liver and human stomach at a 1:100 dilution, with microwave antigen retrieval in 10 mM PBS, pH 7.2, and dot blot analysis has been performed against a mixture of methylation peptides. The product is shipped at 4 °C; on receipt it should be aliquoted and stored at −20 °C, with freeze/thaw cycles avoided. It is intended for research use only and is not for diagnostic or therapeutic applications.
