Anti-Histone H3.3 Antibody

Raised in rabbit, this unconjugated polyclonal reagent from Antibodies.com targets the replacement histone variant Histone H3.3, which substitutes for canonical H3 in replication-independent nucleosome assembly at actively transcribed loci and at sites of active chromatin remodeling.

The antibody is supplied as a 100 µg aliquot and is validated for western blot and immunohistochemistry on human samples, where it is commonly applied to studies of epigenetic regulation, transcriptional activation, and chromatin dynamics. As an IgG preparation reactive with the C-terminal region of H3.3, it allows researchers to track deposition of this variant at promoters, enhancers, and gene bodies independently of DNA replication.

Because H3.3 carries tumor-associated substitutions such as K27M in diffuse midline glioma and G34W in giant cell tumor of bone, the reagent is also used to distinguish wild-type from mutant chromatin marks in fixed tissue sections and whole-cell lysates. For research use only; the vial should be refrigerated in small working aliquots to limit freeze-thaw damage and handled with standard IgG precautions.
